Cheerleaders Hell erzählt die erschütternde Geschichte von vier hübschen Cheerleadern, die nach einer Autopanne mitten im Nirgendwo gestrandet sind. Da sie keinen Handyempfang haben und ein Sturm aufzieht, machen sich zwei von ihnen auf den Weg zu einer nahegelegenen Werkstatt, um Hilfe zu holen – doch dort treffen sie auf Bob, einen unheimlichen 58-Jährigen, der als „Dog Face“ bekannt ist. Bobs perverse Gelüste verwandeln ihre Suche nach Hilfe in einen Albtraum, als er alle vier Mädchen gefangen nimmt und sie in sein abgelegenes Versteck in der Wüste schleppt.

Gefangen in einem Keller voller leerer Hundekäfige werden die Mädchen unvorstellbaren Grausamkeiten ausgesetzt. Bob wählt Jessica, eine umwerfende Blondine, als sein erstes Opfer und zwingt sie, unaussprechliche Qualen zu erdulden. Während die anderen Cheerleaderinnen, Beverly, Anna und Chris, ihre Schreie hören, wird ihnen das wahre Ausmaß ihrer ausweglosen Lage bewusst. Sie sind gefangen, dazu verdammt, für immer Dog Faces Gefangene zu sein, und müssen seinen perversen Forderungen nachkommen – oder mit noch schlimmeren Folgen rechnen.
